64bit? Does cheat engine work in general? Try going to Edit>Settings>Extra and click the top 2 boxes.

What version of cheat engine? Windows 7? If you're using 6.1 and you have windows 7 I have no idea.

To do it manually do this:
1.Make Death Valley Room
2.Scan for 1
3.Start Game
4.Scan for 2
5.Esc-Return to game room
6.Scan for 257
7.Use that address for the steps.

After you copy and paste the addresses like the instructions you can use an auto F5 as long as you keep the game open. No hotkeys required.
