It took 5 hours for me, but it's all luck based.
I was farming on Escherion, which the runes drop rate is 0.1%

I filled a private room on escherion with bots and got it within half an hour. Based on the drop rate and nothing players do can affect the drop rate.

No. -_- I have about ten alts that I log on just for extra help with bosses and stuff. They are all level 60 with Necromancer. That's all you need in an alt.

Le Bot 8.4

Aslo don't try to bot the same account at the same time with multiple windows open. For example Window 1 has earned :40 exp and window 2 has earned 70 exp. When you close both of the window you will have no exp earned since you tried to bot the same account with multiple window.

I think some others who have replied to you are misinformed...
I ran Le Bot for the Runes for 3 weeks and I never received the item.
So I decompiled Le Bot and found that there is a spelling error in the Runes bot set up - it attempts to pick up "Rune Found!" when it should be "Runes Found!"
If you're going to use Le Bot, select "get all drops". The error is present is Le Bot 8.3 and 8.4.

I strangely enough felt there was something wrong. Instead of logging and clearing the item drop list, I actually bothered to manually check myself for whether the runes dropped or not (even then, still no runes after five days of botting ). Well thanks for informing me of a solution venerable.
