I already do, but my wrists and hands tend to hurt after a while, and if i could get it so i can play on an xbox controller, it would give my wrists a break lol.

Okay, so I was looking through the options, and I found what I think you were talking about, but all that does it target them so it shows their health bar, items, etc.
What I'm looking for is basically something that, when activated by button, moves the mouse cursor and hovers over the enemy. I know how to make it so that the minions health bars don't show but the enemy players do, so if it has a pixel check, you can just have it do a search for the enemy health bar color, then, when the button is pressed, the cursor hovers either on the health bar, or right below it (on the enemy themselves).

So i just realized there's no way to customize the movement of the champion via keys, only mouse.

But now, (this would take testing and getting use to) for movement, I would use Xpadder to bind my left joystick to do by right click when pressed, and then moving the joystick would move the cursor, and to move the champion, i would click it, (or use a diff button so i don't destroy my stick) and use an Orbital movement control to move my champion around. from this i can use shift, crtl, and alt modifiers to tell it to auto attack the closest thing to me, or i could quick learn my abilities, etc. only thing left would be this auto aim bot, say i get ganked or something, instead of having to drag my mouse over to the enemy, i could click a button, my mouse will jump to them and i can quick launch an ability and possibly get away, or kill them (lol). and like i said, it would just take a simple pixel search for red (
#CC0000 or
#990000)and a hotkey to activate, then possibly a x,y customization, so, by default, the cursor (center, 0,0) would be on center with the color found, making the target at 0,0. But you'd be able to customize it so that it finds the target color, and when the button is pressed, it puts the target color at say 0,100 (px) that way it puts the cursor (0,0) right below the found target color and putting the cursor on the enemy.
If this doesn't make sense I can draw pictures to discribe it. (hooray paint) xD
