How efficient would it be and is it worth me trying to create it?

Anyways: I assume you want to check if the e-mail is already used.
Sure it is possible, but the amazon api has a limit of requests per minute/hour/day.
If you reach these limit you need to solve a captcha. You could bypass this by using proxies/vpn with diffrent IP's.

I doubt it would be worth your time,since visual basic is not exactly the most powerful of tools
